Human papillomaviruses E6, E7 proteins
Viral early proteins E6 and E7 induce immortalization and transformation in a variety of rodent and human cell types. E7 proteins are shared with other viral oncoproteins, specifically adenovirus E1A and SV40 large T antigen. Provided are recombinant human papillomaviruses type 16 and type 18 E7 proteins, expressed in E. Coli, purified under native condition. E7s are suitable for eliciting cytotoxic T lymphocyte (CTL) response to enhance the gene delivery in vitro. E7 proteins can also be used as antigen to detect the HPV antibody, that raised by HPV infection, in vitro. If the activity is high, clinical examination of cervical cancers is recommended. Production of a soluble E6 oncoprotein is notoriously difficult. Now we have been able to do so. Provided are recombinant human papillomaviruses type 16 wt E6 with his-tag, and type 58 wt E6 with MBP-tag, expressed in E. Coli., purified under native condition. Samples on gel filtration, HPV 16 E6 (his-tag, ~19KD) is eluted later
